Murphy was born on April 3, 1961, and reared in the Bushwick section of Brooklyn, New York City.
His mother, Lillian (Laney), worked as a telephone operator, while his father, Charles Edward Murphy (1940–1969), was a transit police officer who also dabbled in acting.

Murphy was inspired to become a comedian after listening to Richard Pryor’s comedy album That Nigger’s Crazy when he was 15 years old.
Murphy debuted at a talent event at the Roosevelt Youth Center on July 9, 1976, doing an impression of singer Al Green as the song “Let’s Stay Together” played.

Around the end of the 1980s, Murphy had two boys in rapid succession: Eric with then-girlfriend Paulette McNeely and Christian with then-girlfriend Tamara Hood.
Murphy met Nicole Mitchell in 1988 at an NAACP Image Awards presentation and began a long-term relationship with her. They lived together for over two years before marrying on March 18, 1993, in the Grand Ballroom of the Plaza Hotel in New York City. Bria, Myles, Shayne, Zola, and Bella are Murphy and Mitchell’s five children. Mitchell filed for divorce in August 2005, alleging “irreconcilable disagreements.” On April 17, 2006, the divorce was completed.
Murphy began seeing former Spice Girl Melanie Brown after his divorce from Mitchell in 2006, and she fell pregnant and claimed the kid as Murphy’s.
