Murphy was born on April 3, 1961, and reared in the Bushwick section of Brooklyn, New York City.

His mother, Lillian (Laney), worked as a telephone operator, while his father, Charles Edward Murphy (1940–1969), was a transit police officer who also dabbled in acting.

Murphy was eight years old when his single mother fell ill, and he and his older brother Charlie spent a year in foster care.

Murphy has stated in interviews that his stint in foster care influenced the development of his sense of humor.

His mother and stepfather Vernon Lynch, an ice cream plant foreman, reared him and his brother in Roosevelt, New York.

Around the end of the 1980s, Murphy had two boys in rapid succession: Eric with then-girlfriend Paulette McNeely and Christian with then-girlfriend Tamara Hood.

Murphy met Nicole Mitchell in 1988 at an NAACP Image Awards presentation and began a long-term relationship with her. They lived together for over two years before marrying on March 18, 1993, in the Grand Ballroom of the Plaza Hotel in New York City.
